Acetaminophen:N-(4-hydroxyphenyl)acetamide .weight:151.16 g/mol.formula: C8H9NO2. Phenylephrine hydrochloride:3-[(1R)-1-hydroxy-2-(methylamino)ethyl]phenol;hydrochloride,weight: 203.66g/mol,formula:C9H14ClNO2 .

Chlorpheniramine Maleate:(Z)-but-2-enedioic acid;3-(4-chlorophenyl)-N,N-dimethyl-3-pyridin-2-ylpropan-1-amine ,weight:390.9 g/mol,formula:C20H23ClN2O4 .

Pharmacokinetics: 

-Acetaminophen: The plasma half-life:. 1.25-3 hours, elimination: renally, the systemic availability is subject to first – pass metabolism (70 -90%).

-Chlorpheniramine:well absorbed,metabolism:extensive and rapid(gastricmucosa and liver)elimination: in the urine, Plasma half-life:2-4 hours, Terminal elimination half-life (healthy adults and children): 20-24 hours and 10-13 hours, respectively.

-Phenylephrine: it has reduced bioavailability by the oral route due to first – pass metabolism, The pharmacologic effect of phenylephrine is terminated at least in part by uptake of the drug into tissues.

Indications:

For treatment of Cold symptoms,Congestion(nasal and Sinus).

Contraindications:

-This tablet should not be used concurrently with Monoamine Oxidase Inhibitors (MAOIs) or within 2 weeks of discontinuing MAOI therapy.

-It should not be use with alcoholic beverages , head trauma and increased intracranial pressure, Asthma, bronchitis, chronic obstructive pulmonary disease (COPD), emphysema, status asthmaticus, tobacco smoking, hepatic disease, Renal disease, Acute myocardial infarction, angina, arteriosclerosis, cardiac arrhythmias, cardiac disease.

-If there is any sensivity to any of the medications in combination being taken.

Warnings and precautions:

-It should be use with caution in patients with Raynaud's phenomenon or diabetes mellitus, prostatic hypertrophy,closed angle glaucoma.

-Avoid driving or operating machinery until you know how this product will affect you.

-Avoid use in children <12 years of age.

Drug Interactions

CNS depressants or stimulates, alcohol,anticholinergics, medicine for high blood pressure.

Especially allergic reactions,anticholinergic effects(excessive sedation,confusion,dry month,constipation,urinary retention),blood dyscrasias,jaundice,dizziness,palpitations.

Overdosage:

Suspected overdose:Getting emergency help at once.

Dosage and Administration:

Adults:1 or 2 Tablets every 6 hours if needed and don, t use more than 8 Tablets in 24 hours.

Children:it should Prescribe by the physician.

How Supplied:

Griphen ® Adult tablets (325 mg  Acetaminophen , 5 mg Phenylephrine Hcl , 2 mg Chlorpheniramine maleate) are availabele at blisters of 10's in a box of 30 and 100 with leaflet.
